This study used intersectional genetic tools and calcium imaging in rodents to investigate whether brainstem C1 adrenergic neurons promote anxiety-related behaviors, and whether their inhibition could reduce fear and anxiety responses.
Activation of C1 neurons promoted anxiety-related behaviors via the ventrolateral periaqueductal gray (vlPAG); calcium imaging confirmed C1 neuron activation during stress, and targeted inhibition reduced fear responses and prevented anxiety enhancement.
The paper is a preclinical animal study, so direct translation to human anxiety disorders is uncertain. Sample sizes, specific behavioral metrics, and effect magnitudes are not reported in the abstract. The therapeutic relevance of targeting C1 neurons in humans remains untested.
While not yet clinically actionable, these findings identify brainstem C1 adrenergic neurons as a potential therapeutic target for anxiety disorders. Clinicians should watch for future translational work exploring C1-circuit-based interventions.
